Modeling Dengue Immune Responses Mediated by Antibodies: Insights on the Biological Parameters to Describe Dengue Infections

Dengue fever is a viral mosquito-borne disease, significant global health concern, with more than one third of the world population at risk acquiring disease. Caused by 4 antigenically distinct but related virus serotypes, named DENV-1, DENV-2, DENV-3, and DENV-4, infection serotype confers lifelong immunity to that short period temporary cross other serotypes. Severe dengue epidemiologically associated secondary caused heterologous via so-called antibody-dependent enhancement (ADE), an immunological process enhancing new infection. Within-host modeling restricted small number studies so far. With many open questions, understanding immunopathogenesis severe disease during recurrent infections important evaluate impact newly licensed vaccines. In this paper, we revisit framework proposed Sebayang et al. perform detailed sensitivity analysis well-known biological parameters its possible combinations understand existing data sets. Using numerical simulations, investigate features replication, antibody production, clearance over time for three scenarios: primary infection, homologous serotype, serotype. Besides, describing well dynamics as reported in immunology literature, our results provide information on parameter best describe differences viruses.
